I cannot reproduce this using the latest test release (r5701). I tried both the old and the new message view.
(Note that r5701 may have other issues caused by the ongoing transition to a new message view. Older releases can be found here.)

Well that's surprising. It happens immediately for me. Just installed the new public beta, installed Mailmate, started a new compose window, did:
"hello world"
to give it some Markdown to render.
The preview appears on the bottom. I pressed ⌘2 to make it appear on the right, immediate crash.
Is there a way to set this with
defaults write

I failed to mention I didn't try on Big Sur, but the crash reports do not look like it should be Big Sur related. It looks test-release related.
You enable the new message view like this:
defaults write com.freron.MailMate MmNewMessageViewEnabled -bool YES
You disable it again like this:
defaults delete com.freron.MailMate MmNewMessageViewEnabled
Don't do it while MailMate is running.
